Upon homozygous deletion of a small, long-distant enhancer of CEBPA in mice, the expression of the gene is selectively and severely decreased in myeloid progenitors, but not in other CEBPA-expressing tissues. The progenitor cells lost their ability to differentiate into neutrophils and show as myeloblasts. In contrast to bone marrow progenitors of wild-type animals, the enhancer-deleted blasts showed sustained replating capacity using in vitro colony assays.
