Podosomes are dense F-actin structures (green) surrounded by vinculin rings (red) that are formed in monocytes, macrophages, and dendritic cells during migration. Treatment of the human monocytic THP1 cell line with control scrambled short hairpin RNA (shRNA) does not perturb podosome formation. However, cells treated with shRNA against the transcriptional regulator MKL1 have very little F-actin and are unable to form podosomes. See the article by Record et al on page 1527.
