The propeptide of von Willebrand factor (VWFpp) contains the necessary signal or conformation for navigating the regulated secretory pathway. To identify the storage signal within VWFpp, a series of C- or N-terminal VWFpp truncations were expressed in AtT-20 cells and intracellular localization assessed by confocal microscopy. A subset of VWFpp truncations (green) were stored in granules, colocalized (yellow) with endogenous ACTH (red). The VWFpp storage signal appears to be localized within the VWF D2 domain.
