ZHBTc4 embryonic stem cells have been engineered to express Oct4 in a regulated manner. If Oct4 expression is turned off 48 hours after embryoid body (EB) development, brachyury + mesoderm is formed but does not progress to generate hematopoietic cells. However, exogenous expression of SCL (Tal1) bypasses the requirement for Oct4 and allows mesodermal development to proceed to production of red blood cells. See the article by Kong et al on page 60.
