Unresolved replicative stress and common fragile site expression are the main reasons for thrombocytopenia in FANCA-deficient mice. This image shows fluorescently labeled DNA (red) and lamin A/C (green) of polylobulated mouse megakaryocytes demonstrating the accumulation of characteristic nucleoplasmic bridges, a consequence of under-replicated DNA regions in FANCA-deficient cells. The presence of the DNA bridges restrains the endomitotic cycling of the megakaryocytes, affecting their differentiation and normal platelet production in Fanca−/− mice.
