Figure 4. The FLT3 PTK inhibitor PKC412 down-regulates phosphorylation of STAT5 and expression of Bcl-x(L) and RAD51 in FLT3-ITD and FLT3-ITD-TKD Ba/F3 cells. The expression and phosphorylation of STAT5 in Ba/F3 FLT3-WT, FLT3-ITD, and FLT3-ITD-D835Y–expressing cells treated with 100 nM of PKC412 or untreated cells was determined by Western blot analysis in crude lysates using polyclonal pSTAT5 and anti-STAT5 antibodies as well as monoclonal anti–Bcl-x(L) and anti–β-actin (A), and polyclonal anti-RAD51 and monoclonal anti–β-actin (B) antibodies.
