To analyze if a functional TAP is required for the presentation of HLA class I epitopes, autologous DCs were cotransfected with MUC1 and ICP47 RNA (MUC1 alone, ×; ICP47 alone, ♦; both, ▴) and used as targets for the recognition by MUC1-specific CTLs (M1.1 [A] and M1.2 [B]). DCs pulsed with the synthetic cognate MUC1 peptide (▪) or electroporated with irrelevant EGFP RNA (×) were included as controls.
