Figure 2. Overexpressed MtFt is targeted to mitochondria in B9 cells. H1299 cells were transiently transfected with full-length MtFt (48 hours) and the transfected cells were divided into 2 parts. One half of the cells were lysed and the cell extract was used directly for analysis of MtFt expression by Western blotting; the other half of the cells was used to isolate mitochondrial and postmitochondrial fractions. Then, 20 mg of the total cell extracts (T1 and T2, duplicates), 10 mg of the mitochondrial fractions (M1 and M2, duplicates), and 20 mg of the postmitochondrial fractions (C1 and C2, duplicates) were separated by SDS-PAGE and analyzed for the expression of MtFt by Western blotting. The figure shows that in mitochondrial fractions MtFt was concentrated, whereas in postmitochondrial fractions, no MtFt is observed.
