Newborn RAG2-/- γc-/- mice were intraperitoneally injected with human CD34+ hematopoietic stem cells isolated from fetal liver and transduced with the pTRIPΔU3-EF1α p53RNAi lentivirus. Mononuclear cell suspensions from the livers and BM of these mice were stained with various human-specific monoclonal antibodies and analyzed by flow cytometry. Cells positive for human CD45 were gated and further analyzed comparing the GFP+ and GFP- populations Examples of plasmacytoid DCs, myeloid cells, and B cells are shown. Liver and BM cells are stained with antibodies directed to CD123, BDCA2, CD11c, CD14, CD10, CD34, CD19, CD20, IgM, and IgD.
