Figure 6.
Figure 6. The expansion of DC-induced CD44hiCD8+ T cells is impaired in vivo. CD44hiCD8+ and CD44loCD8+ T cells were sorted from the B6 DC-stimulated T-cell cultures at day 5 and labeled with CFSE. Then, 1 × 106 of these B6 DC-stimulated CD44hiCD8+, CD44loCD8+, or unstimulated CCD8+ T cells (CD45.2) were mixed with 5 × 106 B6 T–BM (CD45.1) and transplanted into lethally irradiated B6 mice (3 for each group). Donor CD8+ T cells recovered from the spleens, LNs, and livers of these B6 recipients were analyzed by flow cytometry. (A) The number of donor CD8+ T cells from the spleens, LNs, and livers of B6 mice receiving CD44hiCD8+, CD44loCD8+, or unstimulated CD8+ T cells at days 1, 7, and 14 following transplantation were calculated. *P < .01 (mice receiving CD44hiCD8+ T cells versus mice receiving CD44loCD8+ or unstimulated CD8+ T cells). All values represent means ± SD. (B) The dilution of CFSE fluorescence of donor CD8+ T cells recovered from the spleens, LNs, and livers of these B6 recipients at day 14 following their infusion were analyzed by flow cytometry. Data shown here are gated donor CD8+ T cells, which derive from 3 independent experiments. Numbers in quadrants represent percentages of each cell fraction.
