T1 and T2 populations were harvested on day 6 of culture and restimulated overnight with anti-CD3/anti-CD28–coated beads for potential fasL induction. Effector T cells were then coincubated with 51Cr-labeled L1210/fas targets in Ca2+-free media to inhibit granule exocytosis. The T1 effector population was additionally evaluated in the presence of the fasL blocking agent (rhfasL:fc). (B) Granule-mediated cytolytic function.
