Cells overexpressing BACH2 have increased sensitivity to antileukemic agents. BACH2-overexpressing (no. 67, no. 75) and control (pDL2, pDL3) RAJI clones were seeded (1 × 105/mL) in 96-well plates 1 day prior to experiments. Cells were treated with 0.02 μg/mL MTX (A), 0.01 μg/mL VCR (B), 2 μg/mL VP16 (C), 0.2 μg/mL DNR (D), or 0.1 μg/mL Ara-C (E). After incubation for 24, 48, and 72 hours, MTT (5 mg/mL, 10 μL/well) was added to each well, and the absorbance at 570 nm was measured with the use of a microplate reader.
