Figure 2. Effect of S1P on polyclonal T-cell proliferation. T cells were incubated with 10 ng/mL PMA plus ionomycin (IONO), or 1 μg/mL plated anti-CD3 plus 1 μg/mL anti-CD28, in the absence or the presence of 0.1 to 10 μM S1P for 3 days at 37°C. At this time, 1 μCi (0.037 MBq) [3H]thymidine ([3H]Tdr) was added to each well, and the cultures were incubated for an additional 6 hours before harvesting (A). The same experiment was conducted in serum-free media (SFM) (B). T cells (1 × 106/mL) were treated with 100 ng/mL PTX for 18 hours. These cells were washed 3 times, and then incubated with the various stimuli in the absence or presence of different concentrations of S1P (C). cpm indicates counts per minute.
