(A) A Pax5 cDNA was cloned upstream of the IRES sequence in the parental retroviral vector, MSCV.10 (B) Retroviral expression of Pax5 in NIH-3T3 cells and FACS-sorted myeloid-lineage (Mac-1+Gr-1+) cells isolated from reconstituted animals 12 weeks after transplantation. NIH-3T3 extract was obtained by transduction of cells with control or Pax5-expressing retroviral vectors. One million myeloid-lineage cells were used per lane. Western analysis was done using an anti-Pax5 antibody (Santa Cruz Biotechnology) and ECL.
