Effect of attenuated β-catenin signaling on clonogenicity. / (A) Jurkat and (B) HL-60 cells were transfected with pEGFP (5 μg) and other constructs (10 μg each) as indicated, and GFP-positive cells were isolated by flow cytometry as described in “Materials and methods.” A total of 1000 sorted cells were transferred to 5 mL semisolid medium containing 0.2% agar and plated into 24-well plates (0.3 mL per well). Colonies of 20 or more cells were scored on an inverted Leica microscope after 7 days. At least 4 wells per condition were counted in 3 independent experiments.
