Comparison of dehydration patterns induced by permeabilization to K+ with valinomycin (A), A23187 plus Ca2+ (B), and deoxy (D) versus oxy incubation (C). Cells were suspended at 10% Hct in solution C with 1 mM [Ca2+]o, incubated at 37°C in a tonometer, and at time = 0, 1 mM vanadate was added in the oxy-deoxy conditions (C,D). The cells used for the experiments of panels C and D were from the same donor; those in panels A and B were from different donors. The differences in the control curves of panels A-C reflect the interdonor variation regularly observed in hemolysis curves with sickle discocyte fractions.
