Iron uptake per TfR is greater in C282Y cells than in WT cells.

(A) Iron uptake over time is increased in C282Y cells. Cells (10⁶/mL) were incubated with ⁵⁹Fe-Tf (143 pmol Fe/mL) in fresh medium containing 1% FCS. ⁵⁹Fe uptake by cells was determined at the times shown. Values shown represent means ± SE (n = 3). (B) Iron uptake over a range of exogenous Fe-Tf concentrations is increased in C282Y cells. ⁵⁹Fe uptake was measured over a 270-minute incubation. A representative experiment is shown. In all experiments, cell-surface TfR expression was determined by ¹²⁵I-Tf binding assay at the start of⁵⁹Fe uptake.
