Inhibition of SOCS-3 expression and STAT3 activation by a JAK3 inhibitor.

MyLa 2000 cells were incubated with the indicated inhibitors for 1 hour at 37°C. The JAK3 inhibitor was used at 10, 25, 50, or 100 μg/mL and AG1478 was used at 200 ng/mL. Both inhibitors were diluted in DMSO and the final DMSO concentration in all samples were 1:400. One sample was treated with DMSO 1:400 alone. The cells were subsequently lysed and whole cell lysates of 1 × 10⁶ cells were subjected to Western blotting with the indicated antibodies.
