Comparison of Apaf-1 and procaspases in paired leukemia samples.

Aliquots containing 5 × 10⁵ marrow mononuclear cells harvested before initial chemotherapy (−, odd lanes) and again from the same patient at the time of leukemia regrowth (+, even lanes) were subjected to SDS-PAGE, followed by blotting with reagents that recognize the indicated polypeptide. To provide a standard curve, aliquots containing 5 × 10⁵, 2.5 × 10⁵, 1.25 × 10⁵, and 0.5 × 10⁵ untreated HL-60 cells were loaded onto each blot as shown in Figures 1 through 3. Blots were scanned and quantitated as described in “Materials and methods.”
