CD34+cells (purity, >96%, ≤98%; 105/mL) were incubated in 10% FCS-RPMI, with or without amifostine (8 μg/mL), for 16 hours. Then nuclear extracts were obtained, incubated with a32P-labeled κB oligonucleotide, and analyzed by EMSA. Where indicated, the nuclear extracts were incubated with the32P-labeled κB oligonucleotide in the presence of excess (50×) amounts of unlabeled κB or a different (NF-AT) oligonucleotide.
