Fig. 5. Intravenous injection of TNF- and OSM cooperatively increases mRNA for P- and E-selectin in murine tissues. Each mouse received an intravenous injection of buffer (Control), or of buffer containing 25 ng murine OSM, 3000 U murine TNF-, or a combination of both cytokines. After 1 hour, the mice were killed and the organs were immediately collected for RNA isolation. The levels of mRNA for P-selectin, E-selectin, and β-actin were measured by a competitive RT-PCR method as in Fig 2, except that the primers contained the respective murine cDNA sequences. Shown is representative agarose gel electrophoresis of the PCR products from mouse kidney. The data are representative of 4 independent experiments for control mice and mice receiving OSM, and 2 independent experiments for mice receiving TNF- or the combination of both cytokines.
