PCLT experiments were performed in a reaction mixture containing citrated plasma, various concentrations of mutant 2AP, 1 U/mL thrombin, 20 mmol/L CaCl2, and 200 U/mL UK. Plasma samples were preincubated with mutant 2AP for 10 minutes at 22°C before adding a fresh mixture of UK, thrombin, and CaCl2. Turbidity was monitored as absorbance at 405 nm. PCLT was determined as the midpoint between the highest absorbance and constant lower absorbance.31
