Time course for the formation of 5-oxo-ETE metabolites by resting and activated platelets. Platelets (2 × 10⁸/mL) were incubated with 5-oxo-ETE (2 μmol/L) for various times at 37°C in the absence (A) or presence (B) of thrombin (1 U/mL). 5-HETE (•), 5-oxo-12-HETE (▿; 5o-12h), and 8-trans-5-oxo-12-HETE (▵; 8t-5o-12h), and 5S,12S-diHETE (▪; 5S,12S) were analyzed by precolumn extraction/RP-HPLC as described in Materials and Methods. All values are means ± SE of determinations on platelets from four different donors.
