Figure 2. Defective Ca2+ mobilization following chronic NKG2D stimulation. NK cells were loaded with Indo-1 before incubating with anti-NKG2D antiserum, anti-CD16/32 (2.4G2), anti-NK1.1 (PK136), anti-NKp46 antiserum, or anti-Ly49D (4E5) antibody. Residual B and T cells were excluded by gating on CD19 and CD3-negative cells. Data acquisition was interrupted for cross-linking the bound mAb with goat anti-rat or anti-mouse IgG. Histograms show the cumulative FL5 to FL4 fluorescence ratio over 2.5 minutes whereby the baseline is not included. Numbers depict the percentage of cells with an elevated FL5/FL4 ratio.
