(A) Recruitment of adoptively transferred neutrophils to the lungs in the E coli–induced neutropenia-related pneumonia model. Neutrophils isolated from WT and myeloid-specific PTEN-deficient mice were labeled with CFSE or SNARF-1. Labeled cells were mixed (1:1) as indicated and then injected intravenously (via tail vein) into WT neutropenic mice that had been challenged with 105 CFU of E coli for 2.5 hours. BALF was harvested 2.5 hours after the injection of the cell mixture. (B) The amount of adoptively transferred neutrophils recruited to the lungs was analyzed using a FACSCanto II flow cytometer and FACSDiva software.
