In panels C and D, J16 cells were treated for 30 minutes with 15 μM AQX-016A, 25 μM LY294002, or carrier prior to stimulation with 50 ng/mL of LPS for 15 minutes at 37°C. Cellular lipids were extracted and analyzed for PIP3 (C) and PI-3,4-P2 (D) levels as described in “Materials and methods.” Data are expressed as means (± SEM) and are representative of 3 independent experiments.
