Enhanced AKT activity in CD4⁺CD25⁺ Treg cells restores their capacity to produce IFN-γ, TNF-α, IL-4, and IL-10, but not IL-2. AKT-ER– (or control pCCL-) transduced CD4⁺CD25⁺ Treg-cell or CD4⁺CD25⁻ T-cell lines were activated with αCD3/CD28 mAbs in the presence of either vehicle alone or 4HT (150 nM). Supernatants were collected after 24 hours (A) or 48 hours (B) and assayed for amounts of cytokines by cytometric bead array (CBA).
