Fig. 9. Effect of IL-10 on TNF-RII promoter activity in RAW264.7 cells. RAW264.7 cells were transfected with a 1.8-kb human TNF-RII promoter construct (pGL2-1.8-kb HindIII-luc) using calcium phosphate. Forty hours after transfection, cells were treated with either medium alone (control), IL-10 (10 ng/mL), LPS (100 ng/mL), or LPS + IL-10 for 6 hours at 37°C. Cells were then harvested and cell lysates assayed for luciferase activity. Results are expressed as RLU after normalizing to total protein concentration. Results are representative of 3 similar experiments.
