Monocytes (5 × 106 cells/mL) were pretreated with medium alone or rhIFN-γ (10 ng/mL) for 30 minutes at 37°C and then stimulated with LPS (100 ng/mL) for 0, 1, 2, 4, or 8 hours. At the designated time points, cells were harvested, RNA isolated, and analyzed by Northern blotting using radiolabeled cDNA probes specific for TNF-α and TNF-RII. Similar results were obtained in 2 separate experiments.
