CD24 beads derived from PMNs were stained with MoAb HECA-452 (sialylLex/a) and MoAb CSLEX (sialylLex) followed by phycoerythrin (PE)-conjugated secondary antibodies goat-antirat-IgG or antimouse-IgG, respectively, or biotinylated CD57 MoAb followed by streptavidin-PE. Beads were stained also after treatment with OSGE from P hemolytica to cleave the protein at O-linked glycans. Samples treated with enzyme buffer alone did not show any change (not shown). Note that similar results were obtained with beads coated with CD24 antigen derived from SW2 tumor cells.
