SKT6 cells (2.5 × 105 cells/mL) were cultured in the presence or absence of EPO (±10 U/mL) for the indicated intervals. Lysates then were prepared and were analyzed by Western blotting using antibodies to HCP, Syp, EPOR, Stat5a/b, Stat5a, GATA-1, and murine globins (as indexed in the right margin). For each lane, lysate from 2 × 105 cells was analyzed. For Syp, its correspondence to the upper protein of the two antigens detected was confirmed by the reprobing of stripped blots with a distinct Syp antibody (E.R.S., data not shown).
