Enriched monocyte suspensions (2 × 106 cells/0.1 mL) obtained by centrifugation on a Percoll density gradient were incubated or not with EBV (for 20 minutes) at 37°C, diluted 10-fold with HBSS, and stimulated with fMLP (1 μmol/L for 10 minutes). Samples were subjected to RP-HPLC and fractions containing arachidonic acid were collected for quantitation by mass spectrometry as described in the Materials and Methods. Results (mean ± SD) are from one experiment representative of three separate experiments performed in triplicate.
