TIMP-3 down-regulated the production of IL-12 and TNF-α in DCs. imDCs were stimulated by LPS (1 μg/mL), TIMP-3 (50 ng/mL), or their combinations for maturation. Otherwise, LPS (1 μg/mL) was supplemented to the TIMP-3-knockdowned imDCs (si-TIMP-3) or their corresponding control (si-Ctrl). Two days later, the cell-free supernatants of different cultures were collected and analyzed for IL-1β, IL-6, IL-10, IL-12 p70, and TNF-α production by the Bio-Plex Protein Array system.
