Tregs were seeded at 1 × 105/well in a 48-well plate with irradiated allogeneic DCs (1 × 104), rapamycin (100 ng/mL), IL-2 (10 U/mL), and IL-15 (10 ng/mL). Cells were cultured for 12 days. (A) The total Treg numbers at each time point were calculated by flow cytometry of timed events. The numbers of starting Tregs were normalized to 1 million at time 0.
