Flow cytometry of engrafted human cells in NSG/SGM3 mice. 1-3 × 105 FACS-sorted CD34+ cells from a normal donor (A) and CD34+IL-3Rα+ (B-D) or CD34+IL-3Rα− cells (E-F) from FA-AML patients were injected into the right femurs of sublethally irradiated NSG/SGM3 recipient mice. Six weeks after transplantation, bone marrow cells were isolated and analyzed for the engraftment of human cells using human anti-CD45 antibody (left panels). The right panels show the CD34 compared with the IL-3Rα profiles of the human CD45+ gated population. For each sample, 50 000-100 000 events were analyzed.
