Dasatinib increases MK ploidy in vitro. Progenitor cells isolated from murine BM were treated with 10μM dasatinib along with TPO at the beginning of the differentiation process and 24 hours later. Four days after the addition of TPO, DNA ploidy was analyzed using flow cytometry by staining purified BM-derived mature MKs with propidium iodide and anti–mouse GPIIb-FITC antibody.
