GPVI signaling requires both ITAM tyrosines. DT40 cells were transfected with 2 μg/mL stated FcRγ-chain construct, 2 μg/mL GPVI, along with an NFAT-luciferase reporter gene. Transfected cells were then stimulated with 10 μg/mL collagen for 6 hours at 37°C, after which time the amount of luciferase activity was measured as a readout of signaling. Results were normalized for transfection efficiency and plotted as a percentage of the wild-type response. Error bars represent the geometric mean ± SE of 3 separate experiments.
