Fresh BM from Wt and Nox1 KO mice was fixed in 70% ethanol, washed, stained with CD41-FITC antibody, followed by propidium iodide (PI) staining before analysis on a BD FACScan. (B) MK ploidy profiles from Wt and Nox1 KO mice. BM from Wt and Nox1 KO mice was cultured and stimulated with TPO for 3 days. Cells were then fixed in 70% ethanol, washed, stained with CD41-FITC antibody, followed by PI staining before analysis on a BD FACScan. Each experiment was repeated 6 times.
