Figure 3
Figure 3. Representative Z-series confocal microscopy pictures of the eyes and brains of mice treated with WT BMCs. Transplanted, BMC-derived, GFP-positive cells are seen in green. (A-D) Eye. Bodipy-phalloidin is seen in red. Scale bars represent 50 μm (24×/0.6 NA air objective). GFP-positive cells were observed in the corneal stroma (A) and adjacent to the basal cells of the corneal epithelium (B). Heavy perivascular infiltrates around the limbus were also observed (C). Corneal cystine crystals (D, arrow). (E-H) Brain. Scale bars represent 10 μm (E: 60×/1.4 NA oil objective; F-H: 40×/1.3 NA oil objective). The majority of the GFP-positive cells were observed in association with blood vessels. Small round cells were observed in the lumen, many appearing to be attached to the endothelium. Large flat cells were either fused with or differentiated into smooth muscle cells colocalizing with F-actin seen in red (E, yellow, arrow) or endothelial cells colocalizing with VWF seen in blue (F, white, arrow). A few GFP-positive cells were macrophages (G, white, arrow). Some were glial cells colocalizing with glial fibrillary acidic protein staining seen in red (H, yellow, arrow).
