Figure 3. IFN-α2b suppressed expression of transcription factors regulating late-stage megakaryopoiesis. Human bone marrow–derived CD34+ cells were incubated with 10 ng/mL TPO for 7 days. Large megakaryocytes were enriched by velocity sedimentation, followed by 6 days of incubation with 10 ng/mL TPO and 0, 1, or 10 ng/mL IFN-α2b. Total RNA was extracted from these megakaryocytes. TaqMan Gene Expression Assay was used for real-time PCR. All data were standardized with respective HPRT mRNA levels. Data are means plus or minus SEM of 3 samples.
