p210 level is restored in the rescued SHP-2^Δ/Δ cells transduced by Bcr-Abl. SV40 large T-antigen–immortalized WT, SHP-2^Δ/Δ, and SHP-2–rescued embryonic fibroblasts were transduced with Bcr-Abl or the vector control. (A) Transduced cells were sorted and analyzed with anti–c-Abl IB. The blot was stripped and reprobed with anti–SHP-2 antibody. (B) Sorted cells were seeded into DMEM with 15% FBS and 0.3% low-melting point agarose (0.5 × 10⁴ cells/mL). After 10 days of culture at 37°C in a humidified 5% CO₂ incubator, colonies consisting of more than 50 cells were scored.
