Induction of more potent Th1 and CTL responses by immunization with DIgR2-siRNA-DCs. C57BL/6 mice were immunized once with DIgR2-siRNA adenovirus-infected DCs that were pulsed with ovalbumin proteins and ex vivo matured with LPS (100 ng/mL) for 24 hours before immunization (1 × 10⁶ DCs per mouse). Two weeks later, splenocytes were pooled from 3 mice from each group. (A) IFN-γ production by CD4⁺ T cells after pooled splenocytes were restimulated with OVA-II peptide (1 mg/mL) for 48 hours. (B) Percentages of H2-K^b/OVA tetramer⁺ in the total gated CD8⁺ T-cell population. (C) Percentages of IFN-γ⁺ T cells in the total gated CD8⁺ T-cell population when pooled splenocytes were restimulated with OVA-I peptide (10 mg/mL) for 3 days.
