Nonadherent bone marrow cells from healthy donors were cultured in 10 ng/mL CSF-M and 50 ng/mL RANKL in the presence of CC-4047 (100, 10, and 1 μM), thalidomide (Thal; 100 μM), or DMSO (0.1%). After culturing for 3 and 4 days, total cell lysates were prepared for Western blotting to determine (A) PU.1 and (B) c-fos protein expression. GAPDH and β-actin antibody were used as loading control.
