Morphologic analysis of the adherent cells stably expressing mutant αIIbβ3 to immobilized fibrinogen.

293 cells stably expressing (A) wild-type αIIbβ3, (B) Tyr143HisαIIbβ3, or (C) KO mutant αIIbβ3 were incubated with immobilized fibrinogen (5 μg/mL) at 37°C for 60 minutes in the presence of 50 μM cyclo(RGDfV) (αvβ3-specific antagonist). After gentle washing with PBS, adherent cells were fixed in 3.7% formaldehyde and permeabilized in 0.5% Triton X-100. Cells were stained with rhodamine–phalloidin and analyzed under a fluorescence microscope. Original magnification, × 100.
