Lytic activity of CTLs generated from patient 641. Autologous iDCs were transfected with either CsiRNA (□/○) or iPsiRNA (■/●). After the induction of DC maturation, these DCs were transfected with either MART RNA (A) or tyrosinase RNA (B) and then used to stimulate autologous T cells in culture. After 1 restimulation, lytic activity was determined in duplicate against autologous DCs transfected with TAA RNA (Mart RNA in panel A and tyrosinase RNA in panel B) or GFP (control) RNA, and the autologous melanoma cell line 641 derived from this patient.
