Figure 7. IGF-1 restores SFLLRN- and AYPGKF-induced Akt phosphorylation upon blockade of Gq pathways in platelets. Aspirin-treated, washed human platelets preincubated with 50 nM YM-254890 were stimulated at 37°C for 3 minutes with either SFLLRN (3 μM) or AYPGKF (200 μM) in the absence and presence of 100 nM IGF-1. The reaction was stopped by the addition of 3 × SDS sample buffer. Platelet proteins were separated by SDS-PAGE, Western-blotted, and probed for anti-phospho-Akt (Ser473) or anti-PKCδ (lane loading control) antibody.
