Figure 5. Akt is phosphorylated in response to IGF-1 in platelets. (A) Washed platelets were stimulated with different concentrations of IGF-1 for 3 minutes at 37°C without stirring. (B) Platelets were stimulated simultaneously with 2-MeSADP (100 nM) and different concentrations of IGF-1 for 3 minutes at 37°C. The reaction was stopped by the addition of 3 × SDS sample buffer. Samples were separated by SDS-PAGE, transferred onto polyvinylidene difluoride membranes, and probed with anti-phospho-Akt (Ser473) or anti-PKCδ (lane loading control) antibody.
