Afterward, cells were washed, stained, and analyzed by flow cytometry. Solid histograms represent the matched isotype controls. (B) DCs generated from adherent monocytes by incubation with GM-CSF and IL-4 were exposed to 1 ng/mL bortezomib for 24 hours. LPS (100 ng/mL), TNF-α (20 ng/mL), CD40L (100 ng/mL), or a cocktail of IL-1 (2 ng/mL), IL-6 (100 ng/mL), PGE2 (1 μg/mL), and TNF-α (20 ng/mL) was present in the culture medium during the last 16 hours of incubation. Thereafter, cells were harvested, stained, and analyzed by flow cytometry.
