Total MNCs (20 × 106 cells/mouse) or CD34+ cells (5 × 105 cells/mouse) purified from cord blood samples were incubated for 40 minutes at 37°C in the presence or absence of FucTVI and GDP-fucose. Cells were then washed and injected in sublethally irradiated (400 cGy) NOD/SCID mice. BM was harvested 12 to 16 hours after injection and analyzed for the content of human CD34+ cells or CD34+/CD38lo/neg by FACS. Shown is the mean ± SEM number of total human CD34+ or CD34+/CD38lo/neg cells recovered per femur in 6 independent experiments. The type of cells injected (total MNCs or purified CD34+ cells) and the number of mice per group in each experiment is indicated (total 36 mice). P values compare the mean number of untreated versus FucTVI-treated homed cells after pooling experiments 1 + 2, 3 + 4, and 5 + 6.
