Recognition of target cells expressing CMV antigens by CD8+ CTL clones isolated directly from the blood of CMV-seropositive donors

Autologous fibroblasts were infected with RV798 for 48 hours or AD169 for 16 hours and used as target cells in a chromium release assay. Aliquots of autologous LCLs were infected with each of the vaccinia CMV recombinant viruses for 12 hours. Clones categorized as lysing RV798-infected target cells exhibited greater than 20% lysis of RV798-infected cells and less than 3% lysis of mock-infected cells. Clones categorized as lysing AD169-infected target cells exhibited greater than 10% lysis of AD169-infected cells and less than 3% lysis of mock-infected cells. Clones were categorized as specific for a CMV antigen expressed by a vaccinia recombinant virus if they exhibited greater than 20% lysis of cells infected with that vaccinia recombinant and less than 5% lysis of cells infected with all the other vaccinia recombinant viruses. The effector-to-target cell ratio in all experiments was 10:1, and the data are derived from duplicate experiments with triplicate samples in each assay.
